Objective: 1. To study 131I-iodinated oil equalization in carcinoma target. 2. To quest a method of detecting 131I-iodinated oil quantitatively in three dimension and calculating the dosage in the carcinoma target. 3. To delay the retention of 131I-iodinated oil in carcinoma target. 4. To observe damage effect of 131I-iodinated oil internal radiation. 5. To establish treatment plan system of 131I-iodinated oil internal radiation.Material and Method: 1. We injected 131I-iodinated oil sector-shapely in analog and tumor model by arc-needle, matched with routine straight-needle injection, and evaluate 131I-iodinated oil well-distribution by plane and three-dimensional image of CT and SPECT. 2. Through the way to reconstruct the images of SPECT, we created the relative equation between the average counts in the image and 131I-iodinated oil density. We injected 131I-iodinated oil into intratumor of C6 glioma and SMMC-7721 hepatoma model, by SPECT reconstructed images assess the three-dimensional density in the tumor, and calculate the dosage of the beta portion. 3. ZT-glue (a -Cyano-acrylic acid) is mixed with 131I-iodinated oil. We observed the mixture coagulation in vitro as well as the mixture stagnation and local metabolic equation in mouse local subcutis and in rabbit liver by SPECT in vivo, contrast with pure 131I-iodinated oil. 4. Tumor damage induced by 131I-iodinated oil intraleision radiation in the carcinoma models are recorded through survival time, whole animal body situation, local carcinoma, pathology, electron microscopy, electrophoresis of DNA ladder, cellular smear staining PI and annexin-v -FITC. 5. As a means of time dose factor we compared 131I-iodinated oil intratumor radiation dosage. By means of arc-needle step-by-step sector-shapely, we created mathmatical model according to ball-shape and ellipsoid-shape drug distribution. Wemerge CT image with SPECT image in order to try the possible relation between treatment target image and carcinoma target image. We established standard table that express the relationship of dosage ratio, accumulative dosage with 131I-iodinated oil density.Result: 1. The method of arc-needle injection with 131I-iodinated oil step-by-step sector-shapely improved obviously the drug distribution, comparing with routine linearity injection. 2. SPECT image reconstruction detect 131I-iodinated oil three-dimensional distribution quantitatively. 131I-iodinated oil metabolism equation may obtained by continuous measurements, in order to calculate absorption dose of p particle at target. 3. ZT glue lengthens obviously 131I-iodinated oil retention in mouse local subcutis and rabbit liver which can rise local radiation effect, decrease injury at normal tissue. We recommend 1:4 volume ratio between ZT glue and 13II-iodinated oil. 4. Cell strains of C6 glioma and SMMC-7721 hepatoma showed a radiation dose threshold to 131I-NaI, about 1.052-2.104 gray (Gy) in 48 hours. While compared with routine linearity injection, arc-needle injection 131I-iodinated oil step-by-step sector-shapely through implant C6 glioma and SMMC-7721 hepatoma at subcutis, Rat survival time increased, the body weight kept less descent, their lumps necrose wholely. Pathology, electron microscope, electrophoresis of DNA ladder, cellular smear staining PI and annexin- v find manifestation of cell necrosis and collapse, electron microscope and cellular smear staining PI and annexin- v find apoptotic cells. No difference existed between two strains and two injection methods.Conclusion: Success to obtained well-distributed diffusion for iodinated oil in target, as well as delayed its retention, increased adjustable and effectuation for its internal radiation. The major damage effect at the level of treatment dosage showed cancer necrosis. The results provide new basis to establish 131I-iodinated oil interventional radiotherapy treatment plan system.
