| Rheumatoid arthritis(RA) is a kind of autoimmunity disease which therapy hardly, It's etiological unknown up to now, The Traditional Chinese medicine has potential superiority on the disease, while modern medicine has so much ill effect. Blood benefited and circulating method has so thick theory background and comprehensive clinical verification, but deficit explanation of modern digit. This is emergency problem. Many studies suggested: cellular immunity and apoptosis have important status in pathology of RA. Maybe it has some relationship with stagnation and deficiency of blood syndrome.1ObjectiveThrough observation of experiment rats which administrated on different phases, survey syndrome by BITONG's effect. Study the perhaps law which existed in stagnation and deficiency of blood syndrome; according to the influence of experiment rats' platelet aggregation, cellular immunity, apoptosis, study part modern mechanism of BITONG.2Method2.1Theoretical method: Through survey of literature and summary of tutor's clinical experience, we summarized and analyzed the possibility of stagnation and deficiency of blood syndrome in RA. 2.2Eheoretical study method: With the model of AA rats, They were randomly divided into normal control group(A); model group(B); BQYY group(C); dexamethasone group(D); BITONG heavy dosage group(E); BITONG middle dosage group(F); BITONG light dosage group(G); BITONG metaphase group(H); BITONG latephase group(I). The intervention of BITONG, BQYY and dexamethasone as control drug, we adopted the following technique in our study. (1) every three days, we measure bulk of ankle joint(including feet) by domestic rat feet volume measuring from immune day, and measure body weight every week. (2)The level of ESR,CRP,RF in blood for 30, 45, 60 days, respectively, pathomorphology investigation on ankle joints. (3) The level of TXB2 and 6-keto-PGF1α in blood with Enzyme-linked immunoadsordent assay(ELISA) at same time, and analyzed the correlation with ESR, CRP, RF. (4) The level of IL-4, IFN-γ in blood with ELISA at same time, and analyzed the correlation with TXB2 and 6-keto-PGF1α. (5)Expression of Fas, FasL, Bcl-2 in synovium with immunohistochemical method for 30, 45, 60 days, respectively.3Result3.1 Sigh of AA and analysis of syndrome maybe existed in AAAA rats presenced tacho and delayed hypersensitivity after administration. they had two swelling climaxes on the first and sixteenth day, ankle joint was notable(P<0.01). body weight was normal. by comparison with B group, difference had notable significance(P<0.05), had obvious dose-effect relationship as well. Compared with C group, E and F groups relieved swelling of joint more, difference had striking significance(P<0.01). from the sixteenth day after administration, H group ameliorated valve suitable to F group(P>0.05), better than I group(P<0.01). implied that BITONG prior to BITONGQUEYANGXUEYAO, therapy effect of earlier period and metaphase prior to latephase.3.2The study of perhaps microcosmic marker of AA state of illness, pathological changes, stagnation and deficiency "the pathological lies in collateral, stagnation and deficiency of blood "is principal mechanism of RA, it's microcosmic maybe manifestated: ESR increase fast, CRP step-up, RF step-up(P<0.01). comparied with C group, E, F group improved the marker which reflect state of illness obviously(P<0.01); the effect of H, I groups suitable to G group(P>0.05). we can saw it under microscope: synovial layer of model group increased obviously, synovial cellular proliferated, small vessels increased, pannus formed, much of lymphocyte infiltrated. Synovial cell proliferated lightly in C group rats' synovial, cell layer increased lightly. E, F, H group hadn't seen obvious abnormality, I group had synovial cell proliferated obviously.3.3The correlation study of stagnation and deficiency state , state of illness and apoptosisTXB2 concentration increased obviously in AA rats' blood, and 6-Keto-PGF1αdecreased obviously(P<0.01), by comparison...
