| It is very common that the chronic liver diseases induced by hepatitis B virus(HBV) in China. Hepatocyte apoptosis is an important mechanism involved in liver damage. It is not fully clarified the relationship between HBV and hepatocyte apoptosis. The DNA fragment of two copies of HBV(ayw subtype) genome was inserted into EcoRV site of plasmid pcDNA3, After selection of polymerase chain reaction(PCR) and restriction enzyme analysis, an eukaryotic expression recombinant plasmid (pAKS) containing tandem copies of HBV genome have been constructed. The eukaryotic expression recombinant plasmid (pAKS) was transfected into HepG2 cells by lipofectamine. The HBV genome replicating in liepatoma cell line was identified by detection of ELISA, SOS-PAGE and Western blot analysis. The sensitivity of apoptosis induced by anticancer drug (Methotrexate, MTX) and serum withdrawal was compared among the human hepatoblastoma cell line(HepG2) and , HKS5 cell line with pAKS and HpcD cell line with pcDNA3. The results indicated HKS5 cell had lower sensitivtity of apoptosis than other cells. Therefore, HBV might inhibit hepatocyte apoptosis. |
