Purpose:Occlusion, leading to myocardial ischemia and anoxia or necrosis is the basic pathogenesis of cardiovascular diseases, the incidence rate of coronary atherosclerosis has increased year by year at home and abroad。It is particularly important to carry out the basic and clinical research. We find that physiology, biochemistry and anatomy are very close between miniature pigs and humans. Therefore, this experiment from viewpoint of lipid infiltration and endothelial damage, referring to the characteristics of the Chinese diet, high-calorie, high-fat and a amount of bile salts are fed way for pigs.The combination of mechanical damage of coronary artery intima and high-fat established the model of coronary atherosclerosis and acute myocardial ischemia. To evaluat coronary artery of Bama miniature pig,using the image model evaluation method based on imaging, histology and ECG. The researchers Observated of atherosclerosis related gene changes of AS pathway in Bama miniature pig to explore the biological mechanism of atherosclerosis. The expression changes by detecting atherosclerotic porcine coronary Bama gene chip technology is closely related to the occurrence of AS gene. We will explore the pathological mechanisms of AS To further.Material and method:A Total of ten healthy Chinese Bama minipigs were 6-8 months of age, which were randomly divided into 2 groups:(1) Normal group: basal diet, no running, no balloon injury, n=5;(2)Model group: super heat, high fat diet, running, balloon injury. In the model group, the minipigs were fed with the high Quantity of heat, high fat diet intervention was performed throughout the experiment. In the third week, we performed balloon angioplasty to strain the 1/3 of the middle and long segment of the left anterior descending in minipigs,whose range of movement is(proximal to distal end / far proximal to proximal)about about 5 mm, 5 times in a row, each lasting 30 seconds, between two interval 30 s. In sixth weeks, the model group of Chinese Bama minipigs were run(spleen deficiency) for 19 weeks. 1. Model of instant myocardial ischemia Experiments were carried out for second weeks, Bama miniature pig model group underwent coronary artery balloon compression(balloon / vessel diameter ratio 1-1.3:1);intravascular ultrasound(IVUS):model and the normal group showed intimal smooth, model group the left anterior descending coronary artery distal intimal tear injury at 1/3. Coronary of model group were invented the operate of artery balloon dilatation. Depression of ST segment was significantly down compared with the normal group, the two groups have statistical significance tester(P<0.01) 2.models of atherosclerosis The normal group average area of vascular stenosis rate was(5.2 + 1.36)%, the model group average area stenosis rate was(39.55 + 4.56)%, the difference was statistically significant(P<0.01).the difference of coronary artery area stenosis rate of different methods was not statistically significant(P>0.05); blood lipid changes, compared with the normal group, Bama miniature pig model group TG, TC, HDL, LDL were significantly increased(P<0.01); pathological changes of endometrial lesions in model group, a large number of visible foam cells, foam cell volume increases, the cytoplasm contains a large number of irregular vacuoles, the normal group did not see abnormal meditation. 3. Differential expression of genes of the AS pathway of coronary atherosclerosis in Chinese Bama minipig. There are 25 genes related to stress response.There are 11 genes up-regulated and 3 down in Bama pig coronary genes compared with normal group In the model group.There are 15 genes related to Lipid Transport and Metabolism.There are 5genes up-regulated and5 down in Bama pig coronary genes compared with normal group In the model group.There are 19 genes related to Cell apoptosis. There are 9 genes up-regulated and 2 down in Bama pig coronary genes compared with normal group In the model group. There are 20 genes related to Cell growth and proliferation. There are 10 genes up-regulated and 2 down in Bama pig coronary genes compared with normal group In the model group.Conclusion: 1.Innovation of the ear vein induced anesthesia combined anesthesia breathing full synchronization, greatly reduce the incidence of ventricular fibrillation during operation; 2.We had created a model evaluation method of instant myocardial ischemia of miniature pig, created the body of visual evaluation method in real time; 3. Pioneered the way of extreme heat and high fat and ballon injury of the model of atherosclerosis 4. We established the evaluation method model of atherosclerosis of Chinese Bama swine including dynamic evaluation method of heart electrophysiology and cardiac webcam in vitrobinding pathological. 5. The stage of experiment of Chinese Bama swine coronary artery pathological provided a new way to explore the mechanism of AS through the stress response and lipid transport pathway.
