| The reason and mechanism of abortion are extremely complex. Currently, the majority of domestic and foreign scholars think that the early embryo'loss is close related to immunity condition and the amount of hormone of the maternal-fetal. Medawar has proposed a concept that embryo is a homogeneous transplant of parent to explain the mutual immunity relations between the mother and embryo in the pregnancy in 1953. Afterwards, Wegmann has proposed that pregnant females are susceptible to intracellular pathogens and are biased towards humoral rather than cell-mediated immunity. Since Th1 cytokines compromise pregnancy and Th2 cytokines are produced at the maternal-fetal interface, they hypothesized that these Th2 cytokines inhibited Th1 responses, improved fetal survival but impaired responses against some pathogens. The embryo might lose if the immunity or endocrine hormone functions lose their balance in the pregnancy. Chinese Herb Medicne has been applied to prevent abortion for thousands of years, but the mechanism of which how to prevent abortion that is not completely known, and the effective ingredient of which can prevent abortion is also not known. Moreover, Chinese Herb Medicne does not have the unification national (international) standard, which is one of the important reasons that the Chinese Medicine can't be made to cure and prevent the diseases in world.This study is aimed to investigate the anti-abortive effect and mechanism of Chinese Herb Medicne and its ingredients. Baicalin hydrate and Baicalein are the ingredient of Scutellaria baicalensis Georgi, Quercetin and Bornvl Acetate are the ingredient of Acorus tatarinow ii Schott, which were selected on the basis of Chinese Medicine theory and Pharmacology, have been applied to prevented embryo loss. Scutellaria baicalensis Georgi, Acorus tatarinow ii Schott, Baicalin hydrate, Baicalein, Quercetin and Bornvl Acetate were taken orally for 0.4mL at day 4,5,6, 7 of gestation every mouse, Meantime anti-abortive medicine progesterone (P4) or Pentoxifylline (PXF) was used, then Mifepristone (RU486) or Lipopolysaccharide (LPS) was injected on day 7 of gestation to induce abortion. On day 9 of gestation the mice are sacrificed by cervical dislocation. The rate of abortion, uterus tissue micro- and super-micro changes are observed. The contents of CD4+ and CD8+ T lymphocytes were assayed by immuno-histobiochemistry and computerized imaging technique. The contents of cytokines IFN-γ, IL-2, IL-4, IL-10 and P4 in uterus lysate were assayed by ELISA, and the expression ratio of costimulatory molecule CD80 and CD86 on spleen cells was flowcytometried. The test results are as following:1. The rate of abortion and embryo absorption are increasing respectively with the increasing of the concentration of RU486 and LPS that was taken orally to induce abortion. Then we have a model that per mouse were injected in RU486 for 80μg on day 7 of gestation to induce abortion, and the mice were sacrificed on day 9 of gestation, and the other model that per mouse were injected in LPS for 0.10μg on day 7 of gestation to induce abortion, and the mice were sacrificed on day 9 of gestation.2. Scutellaria baicalensis Georgi and its ingredients: Baicalin, Baicalein that were taken orally from day 4 to day 7 of gestation can inhibit efficiently the abortions caused by RU486, and which rate of abortion and embryo absorption are significantly lower than the group RU486 (p<0.05 or p<0.01), The quantity of CD4+T lymphocyte is extremely lower than the group RU486 (p<0.01), the quantity of CD8+ T lymphocyte quantity are close with group RU486 (p>0.05). The uterus tissue of group RU486 which was observed that had obvious inflammation and damnification at behind of embryo development. Super-micro structure has some changes such as mitochondrion dead, more lysosome, less secrete granules; the groups of Scutellaria baicalensis Georgi and their ingredients were damaged slightly, which groups of expression of CD80 was significantly lower than group RU486 in spleen cell (p<0.05), and the expression of CD86 was significantly higher than group RU486 (p<0.05 or p <0.01). P4 of uterus in group RU486 is higher than other groups because RU486 can block the combination of P4 and progesterone receptor. RU486 descended the contents of IL-2,IL-4,IL-10 just as inhibiting immunity and increased the contents of IFN-γ, however groups of Scutellaria baicalensis Georgi and its ingredients could increase the levels of IL-4,IL-10, just as descending the contents of IFN-γin uterus.3. Scutellaria baicalensis Georgi and its ingredients: Baicalin, Baicalein that were taken orally from day 4 to day 7 of gestation can inhibit the abortions caused by LPS efficiently, and which uterus tissue were damaged slightly than group LPS, the rate of abortion was reduced from 100.0% to 33.3%, 37.5% and 40.0%, the rate of embryo absorption was reduced from 100.0% to 29.4%, 33.3% and 31.6%. The contents of IL-2 in uterus or serum were close in each group (p>0.05). LPS have decreased the contents of IL-4,IL-10 just as inhibiting immunity and increased the contents of IFN-γ, however, groups of Scutellaria baicalensis Georgi and its ingredients could increase the levels of IL-4,IL-10, just as descending the contents of IFN-γin uterus.4. Acorus tatarinow ii Schott and its ingredients: Quercetin, Bornvl Acetate that were given orally from day 4 to day 7 of gestation, can inhibit efficiently the abortions caused by LPS, and which uterus tissue were damaged slightly than group LPS, the rate of abortion were reduced from 100.0% to 50.0%, 60.0%, 30.0% and40.0%, the rate of embryo absorption were reduced from 100.0% to 53.2%, 60.4% 28.6% and 36.7%. Results showed that the quantity of IFN-γ/IL-4 of group LPS was significant higher than group control(p<0.05), and the group of Quercetin + Bornvl Acetate and group Acorus tatarinow ii Schott was close to group of control (p>0.05). In the experiment the content of Th1/Th2 cytokines had close relation with the rate of abortion and embryo absorption, the content of IFN- gamma was higher than other groups, the rate of abortion and embryo was also higher than other groups.The conclusions we drew by analyzing the test results are as following:1. A good experimental model that are mice which were induced to abortion by being injected RU486 that can block the combination of P4 and progesterone receptor and decrease the levels of IL-4,IL-10 just as inhibiting immunity and increases IFN-γlevel in uterus for a model of disease that was caused by the immunity or incretion out of balance when animal was bred. Another good experimental model that are mice which were induced to abortion by being injected LPS that can induce womb to the inflammation and decrease the levels of IL-4,IL-10 just as inhibiting immunity and increases IFN-γlevel in uterus for a model of disease that was caused by the immunity out of balance or inflammation when animal was bred.2. Scutellaria baicalensis Georgi and its ingredients: Baicalin, Baicalein that were given orally from day 4 to day 7 of gestation can inhibit efficiently the abortions caused by RU486 or LPS, the mechanism is that: Scutellaria baicalensis Georgi and its ingredients not only can scavenge oxygen free radical in vivo and have potential effects to prevent organs from oxidative stress damage and have antibacterial and anti-viral effects, but also can regulate the balance of Th1/Th2 to a environment for embryo development.3. Acorus tatarinow ii Schott and its ingredients: Quercetin, Bornvl Acetate that were given orally from day 4 to day 7 of gestation can inhibit efficiently the abortions caused by LPS, the mechanism is that Acorus tatarinow ii Schott and its ingredients not only can scavenge oxygen free radical in vivo and have the potential effects to prevent organs from oxidative stress damage, can soothe the nerves and ease pain and have an antibacterial effects, but also can regulate the balance of Th1/Th2 to a environment for lymphocyte development. |
