Ipomoea batatas is commonly called scammony,sweet potato and batata,which is usually utilized root tuber.The resources of sweet potato stem and leaf(SPSL)are enormous,but most of SPSL are abadoned in our country.In this paper,the extractives from SPSL (ESPSL)of Dadi-1's(ESPSL-1)and Xushu-18's(ESPSL-2)were studied in bacteriostasis, hypoglycemic and anti-inflammatory,then mathematical models were established.The results are as following:1.Chemical constituents in SPSLThere were phenols,tannin,organic acid,alkaloid,steroid,steroid,flavonoids, anthraquinone coumarin and terpenoidm in both SPSL-1 and SPSL-2.Two varieties of SPSL had the same content of protein,total sugar and ash content,but the fat,polysaccharides and crude fiber were distinct These diversities are possibly connected with growing period and condition and so on.2.Preparation of ESPSL-1Ps-1 was extracted by water for twice,each for 4h,in the temperature 80℃and the solid-liquid ratio of 1:25.Then Ps-1 was precipitated by ethanol and deproteined.It contained D-xylose,D-mannose,D-glucose and D-galactose.F-l was extracted by ethanol with the concentration of 70%for once,each for 4h,in the temperature of 80℃and the solid-liquid ratio was 1:30,and separated by absorbent resin.It contained genistein,leteolin,quercetin,formononetin,apigenin and kaempferol. 3# Bacteriostasis of ESPSLThe bacteriostasis of ESPSL which included Ps-1,F-1,the extractives of SPSL which was extracted by systematic solvents(PE-1,SCh-1,SEA-1,SAc-1,SEt-1,SAq-1)and the extractives of F-l which was separated by absorbent resin with gradient of ethanol(0~100%)were researched.ESPSL inhibited the bacillus(Escherichia coli,Staphylococcus aureus and Shiga bacillus)more than mildew,and SAc-1,SEt-1,Ps-1,F-l had favorable bacteriostasis.Medium pH had the major influence of bacteriostasis on Ps-1 and F-1.However,solution pH had less significant effect on Ps-1 and F-1,neutral and meta-acids medium benefited to bacteriostasis. Similarly,temperature has not significant influence.F-l was separated by absorbent resin with 90%of ethanol showed the best bacteriostasis.ESPSL-2 had no visible inhibition for Rhizopus stolonifer and Aspergillus niger.SAc-2, SEt-2,Ps-2 and F-2 displayed better in inhibiting Escherichia coli,Staphylococcus aureus and Shiga bacillu,F-2 was the best.4.Hypoglycemic activity of ESPSLESPSL-1 had no effect of FBG,serum insulin,glycogen and sugar tolerance in normal mouse,which also couldn't impact the diabetogenic mouse's insulin and islet cell tissue morphous.Et-1,Aq-1,Ps-1 and F-1 could significantly or extremely significantly lower the FBG Et-1,SAq-1,F-l,Aq-1 and Ps-1 were able to obviously elevate the glycogen.Ac-1,SAq-1, Aq-1,Et-1,F-l and Ps-1 were capable of raising the level of blood serum SOD.Aq-1,Ps-1 and F-l had ability to cut down the blood serum MDA.Furthermore,Ac-1,Et-1,Aq-1,SEt-1, SAq-1,F-l and Ps-1 also could extremely improve the glucose tolerance of diabetogenic mouse, which still had large deviation comparing with the normal mouse.Besides,when the adding quantity of Et-1,Aq-1,Ps-1 and F-l achieved 300uL that the inhibition ratio ofα-glucosidase could reach to acarbose's 57.2%,44.2%,46.7%and 76.4%.In all of ESPSL-1,F-l had the best hypoglycemic effectEt-2,Aq-2,Ps-2 and F-2 could significantly lower the diabetogenic mouse's FBG.Et-2, Aq-2,Ps-2,SAq-2 and F-2 could significantly elevate the glycogen and blood serum SOD. Aq-2,Ps-2 and F-2 were able to significantly degrade blood serum MDA.When the adding quantity achieved 300μL,the F-2's the inhibition ratio ofα-glucosidase could reach to acarbose's 43.6%.5.Anti-inflammatory effect of ESPSL Tlie ESPSL of Ch-1,SEt-1,SAq-1,F-l,Ps-1 and ESPSL of Ac-1,Et-1,Aq-1 could significantly and extremely inhibite dimethylbenzene-induced swelling degree of ear in mice respectively.Aq-1,F-l and Et-1 also could obviously inhibit dimethylbenzene-induced the increasing of skin capillary permeability in mice.Moreover,in the cotton ball-induced mouse granuloma's inhibitory action,F-l showed significantly and Et-1,Aq-1,SEt-1 showed extremely significant deviation,compared with inflammatory models.In conclusion,tested by three inflammatory models,ESPSL-1 indicated considerable anti-inflammatory effect and F-1 displayed the best.Comparing with the negative control,Et-2,Aq-2 and AC-2 could inhibit swelling degree of ear,and the inhibition ratio of which were 59.9%,51.4%and 47.8%.Et-2 and Aq-2 had significant deviation in cotton-pellet granulomain in mice,the inhibition ratio were 42.6%and 36.9%,but they all had no significant deviation comparing to positive control.6.Diffence of the bioactivities between ESPSL-1 and ESPSL-2The bacteriostasis of Xushu-18 was better than Dadi-1.There was no significant deviation of FBQ liver starch,SOD and MDA in diabetic mouse,as well as inhibitingα-glucosidase.However,Xushu-18 was inferior to Dadi-1 in inhibiting dimethylbenzene-induced the increasing of skin vascular permeability,dimethylbenzene-induced ear swollen and the cotton-pellet granuloma in mice.7.Mathematical model of ESPSL-1 's bioactivities and chemical constituentsThe content of polysaccharides,flavones and chlorogenic acid in ESPSL-1 affected its bioactivities together.In the bacteriostasis,the major was flavones,which had positive correlation with the inhibition on Escherichia coli and Staphylococcus aureus,and the mathematical models were YEscherichia coli=1.210-0.336X+0.021X2 and Ystaphylococcus avreus= -5.485+0.790X-0.025X2.In affecting FBQ glucose tolerance,glycogen andα-glucosidase,it was unable to find which one was more important.In the anti-inflammatory effect,the major was chlorogenic acid,its mathematical models were Yswelling degree of ear=0.279-0.023X+0.001X2 -4.6E-006X3,Yvascular permeability= -0.192+0.069X-0.004X2-7.60E-005X3 and Ygranuloma= -0.001+0.008X-5.05E-006X3.ESPSL had bioacitivities in bacteriostasis,anti-inflammatory and hypoglycemic effect.The bioacitivities associated with variety and the chemical composition of polysaccharides,flavone and chlorogenic acid.There was feasibility and perspective in the exploitation of SPSL in our country.
