Rapeseed is one of the cardinal oil plants in our country and the world. Deodorizer distillate that are produced during processing rapeseed oil has 4-7% Ve and a good resource for Ve. Output of deodorzier distillate is 60-120 thousand ton. This dissertation researched from preparation to bioactivities. The main results are shown as follows:I. This dissertation first researched on extraction Ve from rapeseed deodorizer distillates by supercritical CChwith. cosolvents. Results were as followed:The extracting condition was optimized by Response Surface Methodology Experiment results showed: under the condition of 65C esterified temperature, esterified reaction for 3hr, 0.04 % vitriol acid; 73.7"C hydrolyzed-esterified temperature, hydrolyzed-esterified reaction for 2.1hr, 0.51 % methanol natrium, esterified rate of FFA reached over 90% and hydrolyzed-esterified rate of glycerol ester also reached 90%, loss rate of VEwas 15% or so, Ve content of materials for supercritical fluid extract was about 5.94%, output rate was 3.27%.The supercritical CO2 extracting condition was optimized by orthogonal experiment and single factor experiments results showed: under the condition of 12.0MPa extract pressure, 60C extract temperature, 60C-60C-60C-60C separate temperature field, 15.0kg/hr flow rate, fatty acid methyl esters were extracted, then under the condition of 20.0MPa extract pressure, 40 C extract temperature, 40C-55C-70C-80C separate temperature field, 25.0kg/hr flow rate, Ve were extracted. Ve content of extract reached over 17%, output rate 1.87%.Study on effects of methanol, ethanol, mixed ethanol(methanol : ethanol = 2 :1) as cosolvents on Ve extraction by supercritical CO2 was confirmed. As for Ve content, object extract (VE content were over 40%) weight and Ve quality in object extract were dependent on the cosolvents kind and the quantity of cosolvent. Effects of 4% mixed methanol was best. The effects were as followed: VE contents were improved to 50-55%, extract time was reduced by 33.3%; object extract weight were improved by 31%; extract time was decreased by 28.6%; all VE weight in the object extracr was improved by 55%. 4% mixed ethanol as cosolvent, rapeseed VE content in supercritical extract was 54.17%,output rate was 0.84%.II . It was first that studied the isomers constitutes and identifyied characterictises of rapeseed VE in extracts by supercritical CO2Characters of Ve were confirmed by LC-MS-MS. Ve content in supercritical extract was 54.17%. Ve relative content of each isomers in the extract were showed as follow: a-VE was 12.01 %, B-+r-VE was over 59.29%, 8-VE was about 28.70%.Compared with rapeseed deodorize distillates, polarition degree of rapeseed VE by supercritical CO2 with 4% mixed ehtanol as cosolvent increased to 45.90, acid value, saponification value, heavy metal content (iron, lead and cobalt) and refracting degree were all in the decrease, heavy metal content were below state edible standard. These results showed that characteristics of rapeseed Ve were improved by supercritical extraction.IH.This disseration systemically studied effects of rapeseed Ve on hyperlipidemia in preventive and theraptic experiments. Results showed rapeseed Ve had better effects on preventing and therapying hyperlipidemia.In preventive hyperlipidemia experiments, rapeseed VE decreased the increase weight , blood TC, blood AI wet feces and dry feces of mice, a -VE reduced blood MDA. Rapeseed VE + a -VE reduced blood TG, blood AI, MDA , liver coefficient, liver TC. In therapy hyperlipid experiments, rapeseed VE reduced blood TC, TG, LDL-C, MDA, AI , liver TG and liver coefficient. The mechanism that VE had preventive effects on hypelipidemia was that Y -VE accelerated cholestoral to metabolize from the body.In therapetic hyperlipidemia experients, rapeseed VE had dramatic effects on decrease of blood TG and liver TG of mice. There were no difference between other results. Therefore, rapeseed VE in therapetic hypelipidemia had more better effects than a -Ve, which showed the mechanism that VE...
